Portuguese Abstract: Increased ecstasy use and drug trafficking, in addition to the use of different substances to adulterate and mask the identification tests used by the police, demands the development of new isolation and purification methodologies that may assist in the characterization and elucidation of samples in future seizures. This study, through a partnership between UFRN and the Federal Police, proposed the isolation and characterization of 3,4-methylenedioxymethamphetamine (MDMA) from a sample seized in the state of Rio Grande do Norte, in order to contribute to the obtaining of a substance. that can be used to identify seized material, besides allowing the development and validation of analytical methodology to quantify these compounds. The work presents the comparison between classical column isolation methodologies and the development of a selective analytical method for the determination of MDMA, using gas chromatography coupled to the mass spectrometer (GC-MS). Isolation was compared using classic column chromatography containing Sephadex™ LH-20 (exclusion) and column with Silica Gel 60 (0.063-0.200 mm) (adsorption). The two isolation methodologies used demonstrated efficiency in the seized MDMA purification process. Exclusion chromatography yielded about 5% yield and 100% purity, while adsorption chromatography was shown to produce 10% yield and 100% purity. Identification was performed by comparison with library mass spectra and by retention time. Thus, this study allows the use of MDMA as an analytical standard in PF-RN quantification and identification methodology as also the development of new studies such as process scheduling or increasing yield.
